Abstract
Background and objective: The Iranian strain MJ01 has been reported to produce surfactin, but its genomic potential for use under saline reservoir conditions requires further investigation. This study assessed the genomic placement of MJ01 and compared genes associated with osmotic adaptation and surfactin biosynthesis and regulation in petroleum-associated and reference Bacillus strains.
Methods: Public genome sequences of MJ01, the petroleum-associated strains DM2 and RI4914, the Bacillus spizizenii type strain TU-B-10, and the Bacillus subtilis reference strain 168 were compared. Assembly characteristics, annotation, genome relatedness, and biosynthetic gene clusters were assessed using QUAST, Bakta, FastANI, TYGS, and antiSMASH. BLASTP and targeted nucleotide alignments were used to examine a 35-gene panel covering osmoprotection, potassium homeostasis, stress responses, and surfactin regulation, together with the SrfAA–SrfAD proteins.
Findings: MJ01 shared 99.11% average nucleotide identity with TU-B-10, with a d4 digital DNA–DNA hybridization estimate of 93.4%. Among the 35 target genes, 33 were classified as full-length and conserved in MJ01; opuCD showed near-full query coverage. The SrfAA–SrfAD proteins shared 99.06–99.69% identity with those of TU-B-10 at 100% query coverage. A TTA-to-TGA substitution in the available MJ01 comA sequence predicted a premature stop at codon 53. ComP shared 56.05% protein identity with the TU-B-10 homolog. antiSMASH predicted 12 biosynthetic regions in MJ01, including a region matching the surfactin cluster.
Conclusion: The analyzed gene panel did not support widespread loss of osmoprotection genes in MJ01. Conservation of SrfAA–SrfAD and the predicted comA alteration identify surfactin regulation as a candidate for further investigation. Genome relatedness supports the placement of MJ01 with B. spizizenii. These sequence-based findings do not establish impaired salt tolerance or surfactin production; the comA sequence and its functional consequences require independent validation.
